Part first is concerned with the synthesis and synthetic methods both reported and newly designed and developed, while the second part is about the bioassays evaluation of the synthesized quinazolin-4(3H)-ones. 2-Aryl -4H-3,1-benzoxazin-4-one and methyl-Nacylanthranilates were prepared as starting materials for further transformations into a series of various 2,3-disubstituted quinazolin-4(3H)-ones. Keeping in view the extensive applications of 2-aryl -4H-3,1-benzoxazin-4-one as precursor in the quinazolin-4(3H)-ones chemistry, a new, cheaper andfacile one-pot method for the synthesis of 2-aryl -4H-3,1-benzoxazin-4-one was developed making use of readily available substituted benzoic acids. A faster and high- yielding microwaveassisted, solvent free method was also developed for the synthesis of methyl-2- [(trifluoroacetyl) amino] benzoate. Both the 2-aryl -4H-3,1-benzoxazin-4-one and methyl-N-acylanthranilates synthesized were then utilized for the synthesis of 3-amino, 2-alkyl/aryl quinazolin- 4(3H)-ones and 2,3-disubstituted quinazolin-4(3H)-ones. Consequently, a series of fifteen (19-33) 3-amino- 2-alkyl/aryl substituted quinazolin-4(3H)-ones anda series of twenty seven (46-62) 2,3-disubstituted quinazolin-4(3 H)-ones were synthesized. The necessity of development of new, cheaper and faster methods was realised and as a result, a simpler and less tedious three component one-pot method for the synthesis of ten ( 22-31) 3-amino, 2-aryl quinazolin-4(3H)-ones was developed starting from the available substituted benzoic acid. As a consequenceof the effect of substituents on the reactivity of 2-aryl -4H-3,1- benzoxazin-4-ones, three structurally novel quinazolin-4(3H)-ones (34-36) were synthesized from substituted 2-phenyl-4H-3,1-benzoxazin-4-ones. It was found that phenyl group substituted at position 2 failed to give any product. Reasons for the observed effect onthe reactivity are proposed. Keepingin view the importance of the nature and position of substituents in drug discovery, a series of (37-62) 2, 3-disubstituted quinazolin-4(3H)-ones were synthesized employing three different methods. The effect of substituents on the reactivity and theItem Bioactive Constituents from Mallotus Philipensis and Litsea Monopetala, Synthesis of O-Alkylated Bergenins and Evaluation of Their Biological Activities(INSTITUTE OF CHEMICAL SCIENCES, 2009) Hazrat Amin; Supervisor By: Dr. MohammadArfanFirst ofall I bow my headto the Omniscient, Omnipotent and Omnipresent ALMIGHTY ALLAHwho showered upon meall His blessings throughout my efforts for the completion of this research work. + It gives me a pleasant feeling to acknowledge my inimitable, fabulous and treasured research supervisor Prof. Dr. MohammadArfan, Director of the Institute of Chemical Sciences, University of Peshawar for his keen and continuous interest, valuable guidance and untiring efforts throughout this assignment. Different ligands, ranging from simple non rigid bidentate/tridentate anionic Schiff base ligands to sterically free twisting ligands have been prepared. All these ligands were complexed with transition metals like cobalt, nickel, copper and zinc. The coordination complexes of square planar and distorted tetrahedral geometry were obtained for bidentate anionic Schiff base ligands and octahedral for tridentate Schiff base ligands. Characterization of all the Schiff base ligands were done using various spectroscopic and analytical methods including 1H, 13C{1H}-NMR, infrared spectroscopic method, ES+-MS, elemental analyses and conductance studies. Apart from it Schiff base ligands like 2-[(E)-(quinolin-3-ylimino)methyl]phenol (H-QMP), [1-({[(Z)-(2-hydroxynaphthalen-1-yl)methylidene]amino}methyl)cyclohexyl]acetic acid (H-HHMAC) and 2-(2-hydroxyphenyl)-3-{[(E)-(2-hydroxyphenyl)methylidene]amino}-2,3-dihydroquinazolin-4(1H)-one (H-HHAQ) were recrystallized from concentrated THF solution in Pc, P1 and C2221 space groups respectively. All the synthesized transition metal complexes of the representative Schiff base ligands were also characterized by elemental analyses, ES+-MS, IR and UV Visible spectroscopic studies, magnetic susceptibilities and molar conductance studies. The metal complexes like 1 and 2 were recrystallized by slow diffusion of diethyl ether into the concentrated solution of metal complexes in CH2Cl2 in P21/c space group having trans square planar arrangement of ligand around the metal centers.Item 16_Effects of Thyroid Profile Changes on the Biochemical Markers of Bone, Liver and Renal Function in Non-pregnant Women of Khyber Pakhtunkhwa Pakistan(INSTITUTE OF CHEMICAL SCIENCES, 2012) Mohammad Yousaf; Supervisor By: Dr. JASMIN SHAHThis hospital based cross sectional study was conducted from 01 March 2014 to 31 March 2015 in Khyber Teaching Hospital (KTH), a tertiary care hospital in Khyber Pakhtunkhawa (KP) province of Northern Pakistan. The study group comprises of 259 non pregnant women in the age group of 18-75 years. Informed consent was sought from each patient and all human dignity was respected throughout the study period in accordance with the international norms involving human as an experimental subjects. 5 mL of fresh venous blood (fasting sample) was taken from each patient. It was divided into two portions- one portion was used for the determination of thyroid profile markers by Elisa methods and the other portion was used for the determination of serum alkaline phosphatase, calcium, Zinc, urea, creatinine, SGPT and SGOT level on autoanalyser (Erbamannhein chemistry autoanalyser, Germany) by using Erba kits and Standard protocols. After the determination of thyroid profile status, patients having normal thyroid profile (n=54, TSH≤6.0, normal T3 and T4 levels) were taken as controls. Peace and blessing be upon Prophet Muhammad SAW., his families, his relatives and all his followers. First and foremost, I would like to express my sincere gratitude to my teacher and supervisor Prof. Dr. Ikhtiar Khan for his patience, motivation, enthusiasm, and immense knowledge. It has been an honor to be his student. I appreciate all his contributions of time and ideas to make my Ph.D. experience productive and stimulating. The joy and enthusiasm he has for research work was contagious and motivational for me. He has contributed immensely to my personal and professional grooming at this Institute. I owe my deepest gratitude to my co-supervisor Dr. Abid Mahmood for devoting his precious time, providing numerous ideas, valuable suggestions and research facilities at Barani Agriculture Research Institution, Chakwal, which indeed helped to improve quality of this research work. Without his help and guidance, this work would not have been possible. I am truly indebted and thankful to my foreign supervisor Dr. Barry Lomax, University of Nottingham, for his support and guidance during my six month research at University of Nottingham, United Kingdom.
