BIOTECHNOLOGICAL APPROACHES FOR IMPROVEMENT OF POTATO (SOLANUM TUBEROSUM L.)

Abstract
Shoot cultures were obtained by culturing the buds from disease free healthy
potatoes of the cultivars Desiree and Ultimus, on both 4 and full strength Murashige and
Skoog (MS)(1962) salt solution. Sprouting of the buds in both the cultivars was observed
on the 3“ day of culture on 4 MS medium that grew into single leafy shoots, within 3
weeks, whereas on full MS medium sprouting of the buds in both the cultivars was
delayed and occurred after the second week of culture. Moreover growth ofthe plantlets
was also very slow on this medium. Nodal segments from these plantlets were then
cultured on micropropagation media.
The micropropagation medium consisted of % MS with 4% sucrose and
supplemented with varying concentrations of NAA (0.1, 0.Smg/l), BAP (0.1, 0.5mg/1)
GAS (0.2, 0.4, 0.6, 0.8mg/l) either alone or in combination. The experiments were carried
out in two different day lengths (8 and 16hrs photoperiod). Best multiple shoot formation
was observed on medium supplemented with 0.1mg/l NAA in combination with 0.Smg/1
BAP and 0.4mg/l GAin 16hrs photoperiod. The multiple shoots thus obtained were
further subjected to different tuberization media. The tuberization media consisted of
varying concentrations of different carbon sources (sucrose and glucose); different
growth regulators such as BAP,Kinetin, Adenine sulphate (AS), IAA, Coumarin, ABA,
GAs, Chlorocholine Chloride.
